The Corning Transwell Snapwell has become a mainstay for growing epithelial cells. Its large area (1.12 cm2) and shallow depth are ideal for transport and electrophysiological research in the EasyMount Ussing chamber. The P2302 EasyMount Slider is designed to securely hold these inserts without stress on the membrane and to position the membrane perfectly between the two halves of the P2300 Ussing chamber.

The mounting procedures for placing the Snapwell into the P2302 slider and then into the chamber are both easy and fast. The Snapwell is removed from the culture dish, separated from its holder, drained of media, and placed with its basal side against the red O-ring in the P2302 slider. Wipe the end and sides of the slider to remove any saline and “slide” the Snapwell+P2302 in the slot between the two chamber halves until it reaches the hard stop. Turn the thumbscrew on the chamber to gently squeeze the chamber halves together. Only light pressure between the chamber halves and the slider is required to make a watertight seal. Excessive pressure can distort the culture membrane and alter the electrical parameters. The procedure for mounting the tissue takes less than 30 seconds to accomplish. This system is ideal for transport and electrophysiological measurements. The P2302 enables effortless transfer of confluent cell layers directly into EasyMount Ussing Chambers, providing researchers with a streamlined, effective solution for high-quality results.

P2302 EasyMount Ussing Chamber Slider for Snapwell Cell Culture Insert

The EasyMount Ussing chamber together with the P2302 slider and Snapwell cell culture insert have enabled many scientific advances particularly towards understanding genetic transport defects in cystic fibrosis. The ease and rapidity of mounting these inserts in the EasyMount Ussing chamber have aided drug discovery by increasing the number of experiments that can be run in a single day. 
The P2302 Snapwell inserts are a modified Transwell culture insert that contain a 12 mm diameter tissue culture-treated membrane in a detachable ring. The inserts are made for transport and electrophysiological studies. Once cells are grown to confluence, this ring-supported membrane can be placed into our P2300 EasyMount Ussing Chambers.

1. The tissue sample

Well established cell culture procedures may be used to seed and grow the specific cell type to be studied. The minimal requirement for measurement of transepithelial transport is that the cells grow to confluency in the Snapwell insert. Other factors such as stage of differentiation, days in culture, pretreatment conditions, etc. are established by the investigator.

2. Mounting the Sample:

Once the chamber system has been setup, appropriate solution temperatures have been attained, and electrode offsets and fluid resistance compensation have been adjusted, the cell culture sample can be inserted into the chamber.

A. The first step is to remove the airline from the top of the EasyMount chamber.

B. Remove the solutions from both halves of the EasyMount chamber using either the FN15 ball needle attached to a syringe or to a vacuum tube.

C. Loosen the thumbscrew at least 3 turns and use the slider as a lever to separate the chamber halves. Note: It is important that the opening between the chamber halves is sufficient that slider can be removed or inserted without rubbing on the O-rings on each chamber half.

D. Remove the slider and blank insert used for setting up the chamber.

E. Using forceps and a KimWipe or other tissue paper, dry the surfaces between the chamber halves.

F. Select the sample to be studied and remove from culture tray. Gently lift the Snapwell from its hangar.

G. Tilt the Snapwell on edge against tissue paper to drain the media from the Snapwell. Rinse and repeat with physiologic buffer if needed to remove media.

H. Place the Snapwell into the P2302 holder with the basal side towards the red O-ring.

I. Wipe the sides and end of the slider to remove liquids. The purpose of this is to prevent electrical current from finding a pathway from one chamber to another chamber (crosstalk) via the aluminum heat block. This would cause an error in measured current in both chambers.

J. Insert the P2302 + Snapwell between the chamber halves until it touches the heat block.

K. Tighten the thumbscrew to gently squeeze the slider + Snapwell.

L. Refill the chamber halves with solutions of the correct temperature.

M. Reinsert the airline fittings into the chamber halves.
