The P2302 Snapwell inserts are a modified Transwell culture insert that contains a 12 mm diameter tissue culture-treated membrane supported by a detachable ring. The inserts are made for transport and electrophysiological studies. Once cells are grown to confluence, this ring-supported membrane slider can be placed into our P2300 EasyMount Ussing Chambers.
Instructions on how to use the P2302 EasyMount Sliders with Snapwell inserts:
1. Preparation of Snapwell Inserts
Cell Seeding and Culture: First, prepare your Snapwell inserts by seeding them with the cell type you want to study. Ensure they are cultured until they reach the desired confluency or differentiation stage.
Hydration and Rinse: Rinse the Snapwell inserts with physiological solution to remove any residual media, which could interfere with measurements.
2. Preparing the EasyMount System
Equipment Check: Ensure the EasyMount Ussing chamber system is fully assembled, and all components, including the sliders, electrodes, and chamber halves, are clean and in working order. Each item needs to be clean and dry (no residual fluid on them).
Dry Chambers & Heatblock: Dry the chambers center lower shelf and surrounding areas from fluid that might be left over from a cleaning or previous experiment. Important: Ensure that it is dry on the rear of the chamber that touches the heat block to prevent "crosstalk".
3. Loading the Snapwell Insert into the EasyMount Slider
Snapwell Placement: Take a Snapwell insert and carefully place it into the P2302 EasyMount Slider. The insert should fit securely in the circular opening of the slider without any gaps, ensuring a tight seal.
Alignment: Make sure that the membrane of the Snapwell insert is centered in the slider window, as proper alignment is essential for accurate readings.
4. Mounting the P2302 Slider in the Ussing Chamber
Slider Insertion: Insert the P2302 EasyMount Slider (now holding the Snapwell insert) into the Ussing chamber housing. The slider should easily slide into place, with the membrane positioned between the two chambers.
Chamber Locking: Lock the slider in place using the locks and thumbscrews on the sides of the craddle, to ensure a secure and leak-proof seal between the two chamber halves. This creates isolated compartments for the apical and basolateral sides of the insert membrane.
5. Adding Electrodes and Setting Up the System
Insert Electrodes: Place the voltage-sensing and current-passing electrodes into the respective ports of the EasyMount ussing chamber. Check the electrode placement to ensure proper function.
Connect to Voltage/ Current Clamp: Connect the electrode setup to the DM MC6, and plug DM MC6 into your Voltage Current Clamp.
Fill Chambers with Solution: Fill the chambers with the appropriate physiological buffer solution. The buffer choice depends on your experimental requirements, but it should be matched to the needs of the cells or tissue in the Snapwell inserts.
6. Begin The Experiment’s Measurements
Baseline Check: Begin by recording baseline values to ensure that the system is equilibrated. Check for any leaks or unwanted currents, which can distort measurements.
Start Experiment: Apply the experimental conditions, such as specific ion gradients, agonists, or inhibitors, to observe changes in ion transport or permeability across the cell layer.
7. Post-Experiment Clean-Up
Remove and Clean Components: After the experiment, disassemble the chambers, remove the Snapwell insert, and clean the EasyMount Slider and Ussing chamber components according to lab protocols to prevent contamination.
